Synthesis of Phosphorothioate Oligo
Phosphorothioate Oligonucletiodes is used for experiment of antisense which suppresses expression of target genes specifically. This is the DNA Oligos which phosphorothioated all phosphate group existing at the binding sites between ordinary nucleotides. This binding is called phosphorothioate binding. Structures of phosphorothoate DNA Oligos have resistence properties against almost all exonuclease and endonuclease activities, and they are used for experiment of antisense due to their stability of structures.
  1. Caution for designing
    There are various designs of phosphorotioate DNA Oligos There are complete-type phosphorothioate(All phosphate groups are phosphorothioate) and chimera phosphorothioate( a part of phosphate groups is phosphorothioate) and their proximity of translation start site and regions including borderlines of exon and intron are used.

2'-O-Me-RNA, 2'-F-RNA
It is reported stability against nuclease is upgraded compared to unmodified siRNA by inserting 2'-O-Me into siRNA.
Also, it is said that 2'-F is more stable than 2'-O-Me with double-stranded chains.
If you use it for antisense, it is necessary to phosphorothioation due to nuclease susceptibility.
